چکیده
Currently much attention is focused on glutathione S transferase (GST)-induced suppression of apoptosis. The objective of our studies was therefore to see if GST isoenzymes rescue photoreceptors in retinal explants from rd1/rd1 mice, in which photoreceptors degenerate rapidly. Eyes from C3H rd1/rd1 and / mice were collected at various time points between postnatal day (PN) 2 and PN28. Localization and content of -GST and -GST was investigated by immunofluorescence and semi-quantitative Western blot analysis, respectively. In addition, PN2 and PN7 retinal explants were cultured till PN28, during which they were treated with 10 ng/ml -GST or -GST. The spatiotemporal expression of both GST isoforms was closely similar: early presence in ganglion cell layer after which staining became restricted to Müller cells (particularly in the endfeet) and horizontal cell fibers in both rd1/rd1 and / . Doublets of -GST and -GST were detected by Western blot analysis. Densitometry of these bands indicated steady reduction of -GST content in rd1/rd1 retina starting from the second postnatal week. When -GST and -GST were added exogenously to rd1/rd1 explants, photoreceptor rescue was produced that was more prominent in PN2 than in PN7 explants and more effective by -GST than -GST. We propose that -GST neuroprotection is mediated by reduction of tissue oxidative stress. © 2005 Published by Elsevier Ltd on behalf of IBRO.

متن کاملDifferential modification of phosducin protein in degenerating rd1 retina is associated with constitutively active Ca2+/calmodulin kinase II in rod outer segments.
Retinitis pigmentosa comprises a heterogeneous group of incurable progressive blinding diseases with unknown pathogenic mechanisms. The retinal degeneration 1 (rd1) mouse is a retinitis pigmentosa model that carries a mutation in a rod photoreceptor-specific phosphodiesterase gene, leading to rapid degeneration of these cells. Elucidation of the molecular differences between rd1 and healthy ret...
متن کاملEvidence for glutamate-mediated excitotoxic mechanisms during photoreceptor degeneration in the rd1 mouse retina.
PURPOSE Kinetic studies of photoreceptor cell death in the retinal degeneration (rd1) mouse model suggest that photoreceptor degeneration could result from cumulative damage. Since alterations in glutamate metabolism have been described in different models of retinitis pigmentosa, we investigated in the present work whether changes in glutamate turnover occur in the degenerating rd1 retina and ...
متن کاملPARP1 Gene Knock-Out Increases Resistance to Retinal Degeneration without Affecting Retinal Function
Retinitis pigmentosa (RP) is a group of inherited neurodegenerative diseases affecting photoreceptors and causing blindness in humans. Previously, excessive activation of enzymes belonging to the poly-ADP-ribose polymerase (PARP) group was shown to be involved in photoreceptor degeneration in the human homologous rd1 mouse model for RP. Since there are at least 16 different PARP isoforms, we in...
متن کاملApoptosis in the murine rd1 retinal degeneration is predominantly p53-independent.
PURPOSE To determine if p53 mediates apoptosis in photoreceptors of retinal degeneration, rd1, mice. METHODS The rd1/rd1 mice were interbred with p53 null mice to generate p53-/- rd1/rd1 and p53+/+ rd1/rd1 mice. Rates of loss and incidence of apoptosis in rod photoreceptors were analyzed at appropriate ages (postnatal days 12, 14 and 16). RESULTS The extent and kinetics of photoreceptor cel...
